單細胞測序在探索生物過程、疾病機理等方面展現了前所未有的精度，通過對單細胞進行 DNA 和 RNA-seq 我們得以高精度的了解單個細胞水平的體細胞突變情況，并且對于一個樣本中細胞類型的組成和解析獲得空前的認識。單細胞RNA-seq目前應用較多的是基于mRNA水平測序，如10× genomics，Smart-seq等，但對于單細胞水平全轉錄組目前市面上的研究的方法較少。NuGEN公司推出一款Solo RNA-seq試劑盒可應用于單細胞全轉錄組研究，下面小編就給大家介紹這款試劑盒。

  Ovation SoLo RNA-Seq System

  試劑盒特點：

  1-500個細胞，10pg-10ng total RNA起始;

  Anydeplete技術去除rRNA，產物包含mRNA及LncRNA;

  適用于單細胞，微量RNA，FFPE樣本，外泌體樣本等;

  有人，鼠及果蠅試劑盒，其他已知rRNA序列的物種可定制去除rRNA;

  產物為適用于illumina平臺測序的文庫。

  Anydeplete rRNA去除原理：

  隨機引物進行一鏈合成，再進行二鏈合成，酶打斷補平后加接頭，變性形成單鏈文庫，加入探針引物，探針能夠特異性與rRNA文庫結合，延伸形成雙鏈結構，通過酶將rRNA文庫雙鏈降解，而mRNA和LncRNA文庫保留下來，再通過接頭引物擴增，得到的就是去除rRNA的文庫。
